飛**械工程師凱莉普雷﹝茱蒂佛斯特飾演﹞帶著自己的女兒茱莉亞搭乘自己親手設計的最新型飛機從德國柏林返回美國紐約。上機之後，母女兩人一起陷入沉睡，凱莉一覺醒來發現茱莉亞不在身邊，於是開始在機艙裡尋找自己的女兒。遍尋不著女兒之後，凱莉陷入情緒失控狀態，要求機長瑞奇﹝西恩賓飾演﹞以及同型機組人員以地毯式搜尋方式找遍機上茱莉亞可能藏匿之處仍然找不到女兒下落。凱莉懷疑自己的女兒遭到壞人綁架，但是機組人員在與柏林地面聯絡之後，卻傳回茱莉亞其實早在七天前死亡，並沒有隨著凱莉登機的消息。長期服用安眠藥的凱莉坦承在飛機出發前，曾經服用安眠藥，因此更令人懷疑他是否神智清楚。但是班機上一共有三百多名乘客，包括幾名神情詭異的中東人，凱莉發現這群人彷彿曾經在他家附近出現過，這讓茱莉亞的失蹤更顯得詭異。
所有飛機上的乘客都把凱莉當成是剛剛失去女兒，頭腦不清楚的母親，沒有人願意對他伸出援手，這時出現了一位空中便衣武警─吉安卡森，在911事件後，空中便衣武警便成為每班飛機上唯一配槍的治安維持人員。吉安卡森向凱莉伸出援手，除了安撫凱莉之外，也努力幫助凱莉尋找她的女兒。事情到底會如何發展？凱莉六歲的女兒茱莉亞怎麼可能在四萬英尺的高空憑空消失？凱莉到底應該相信自己還是別人？

Kyle Pratt (Jodie Foster) who is aircraft propulsion engineer  boarding an airplane she helped design with her daughter Julia from Germany to New York City. After boarded airplane,Kyle and Julia sat down for a nap. When Kyle woke up only to find that Julia is missing, she started to looke for her daughter through the plan. Kyle becomes increasingly frantic as she searches the plane for her daughter without success. She demands captain of the plane (Sean Bean) and whole crews that every inch of the  plane be searched. And when each search comes up empty.Kyle started to believe that Julia might have been kidnaped. But when the crews checked with the ground crews in Berlin and find no record of her daughter ever being on board with Kyle. Kyle admitted that she has been under treatment of medications, and took some sleeping pills before on board. All three hundred or more passengers, including several mideast misteious person who Kyle thought were wondering around her house,are suspects.  Everyone on board of the plane thought she is just losing her mind from grief of the loss of her daughter. Nobody would like help her out. At this moment a somewhat friendly air marshal (played by Peter Sarsgaard), after 911 incident, air marshal becomes the only official carries gun on airplane,who offers to help Kyle with the search. He tried to calm Kyle down and did his best to help Kyle looking for her daughter .How would the incident develope. How could Kyle's six year old daughter Julia vanishes in the middle of a flight above 40,000 of feet in the air?  Should Kyle believe herself or others?
