The first satellite ever launched was Sputnik, but I wouldn't call that a capsule. There were unmanned test flights of the Vostok capsule before Yuri Gagarin flew in one to become the first man in space. That was a capsule.

There were certainly suborbital space capsules before Sputnik--some with live animals..
