What causes people to song off key and is there any cure?

It's largely biological (specifically neurobiological). In a normally funcioning sense of hearing, sound stimulates the receptive organs in the ear. This stimulation is converted into electronic signals that are sent to the brain.
The brain interprets these signals automatically (very subjectively, I might add).
The concious interprets the interpretation of the signals. Some people have trained their concious mind to be better at interpreting audio data. They can more accurately determine the acutal meaning of the original data that entered the ear.
If someone sings, they make their vocal chords vibrate at a frequency they believe matches the pitch they are hearing. If they misinterpreted the pitch they heard, the tone they sing will generally not match up properly, thus creating off-key singing.

The best cure is usually practice. Practice listening to music and singing along with it. Record yourself and listen to the recordings. Figure out where you are messing up and how to change that. The big exception is people who have suffered hearing loss. They can help their singing abilities by improving their hearing.

Booze makes people sing off key, trust me I ran a kareoke show and played mostly night clubs & bars. The cure, record them and give them the tape.Most of them dont come back.
